define Restriction Endonucleases
cut double stranded DNA at specific sequences. Used in cloning and some diagnostic assays (e.g., RFLP).
define DNA polymerase
polymerizes a new DNA molecule that is complementary to an existing template strand. It has many uses in molecular biology.
define heat stable DNA polymerases
a class of DNA polymerases, isolated from thermophilic bacteria, that remain active at high temperatures. Taq DNA polymerase (from Thermus aquaticus) is the most common example. Uses include PCR and cycle sequencing.
define DNA ligases
forms a phosphodiester linkage between the 5’-phosphate and the 3’-hydroxyl of adjacent nucleosides on “nicked” DNA. Used in cloning.
