Enzyme Kinetics Flashcards
What is enzyme kinetics?
the study of the rate of formation of products from substrates in the presence of an enzyme
What is the reaction rate?
V = amount of product formed per unit time, in moles per second (mol/s)
What is the reaction rate dependent on?
concentration of the substrate and enzyme
What does it mean for an enzyme to be saturated?
Vmax = so much substrate that the active site is continuously occupied and adding more substrate does not increase the rate of the reaction
What is the Michaelis constant?
Km = substrate concentration at which the reaction velocity is half of its maximum
What does it mean for a substrate to bind cooperatively?
binding of one substrate to one subunit modulates the affinity of other subunits for substrate
What are the two types of cooperativity?
positive
negative
What is positive cooperativity?
binding of a substrate to one subunit increases the affinity of the other subunits substrate
What is negative cooperativity?
binding of a substrate to one subunit reduces the affinity of the other subunits for substrate
What type of enzymes does cooperativity not apply to?
catalytic enzymes
