Q

how does substrate concentration effect enzyme reaction.

A

increase in substrate level at fixed enzyme concentration will allow one to see an increase in rate of reaction up to a point where maximum enzyme-substrate complexes occur thus the remaining substrate has to wait until a an active site is free for binding. at this point the reaction rate is constant

Q

what is the common IU unit used in enzyme reaction

A

Rate is the number of conversions of substrate per unit time.
Common IU unit is the amount of enzyme which catalyses conversion of 1um of substrate per minute (1IU = 1𝜇mol/min⁡)

Q

what unit was specific activities of enzyme expressed as ?

A

The specific activity of an enzyme is expressed as 𝜇mol/min/mg of protein or IU/mg

Q

when is Enzyme velocity measured? why is it measured t this point?

A

Enzyme velocities (V0) are measured at time 0 (or before 10% of substrate is used up)

Prevents observed effects by feedback inhibition or reversible reactions

Q

how does the enzyme concentration affect rate of reaction?

A

same reaction rate trend is seen as if your changing the substrate concentration.
as all substrate will be in a reaction with the use use of enzymes. the excess enzymes left do not react as no remaining substrate present.

Q

how does pH and temperature effect enzyme reaction?

A

optimum graph can be seen for both factors. the rate of reaction increases as the temperature increases , however at a certain high temp, denaturing occurs which causes rate of reaction to decrease. same trend for pH. the reaction rate decreases due to bonds becoming distorted and changed withing the active site, thus rendering the enzyme non-functional.
