Enzyme Flashcards
Lock + key
Shape of substrate + enzyme active site are COMPLEMENTARY , enzyme is specific
induced fit
Enzyme active site changes shape to ACCOMODATE substrate once substrate binds
- conformational change
Effect of high temp
Increase kinetic energy , more successful collisions , more ESC complexes form. Too high temp - Faster initial rate then DENATURES ( so may not reacted w all substrate) , affects bonds in TERTIARY structure , changes shape of active site - prevents substrate binding
Low temp effects reversible - fewer successful collisions
Things to improve experiment
-control PH, TEMP, CONC. , vol of solutions
- temp intervals closer tg more accurate estimate of OPTIMUM temp
Control group
Take samples at more frequent intervals
Use coloritometer
Use PH probe - numerical value
Unit + formula flr rate
1/time
Mm 2/s-1
Unpaired t test
Compares means of two data sets
What does standard deviation show
Shows spread of data AROUND mean
Small SD- little variation in repeats , high repeatability
As conc increases, SD increases , repeatability decreases
Enzyme conc effect
As increases , rate increases , MORE ESC formed, more active sites available , conc of substrate remain the same , rate levels off substrate conc is limiting
Non- competitive inhibitor evfect
Bind to allosteric site, changes shape of active site , substrate can no longer bind to it
Cofactor
Inorganic ion which helps
CL- for amylase
ZN2+ - carbonic anyhdrase
Competitive inhibitor
Competes w substrate to bind to active site, binds to active site + prevents substrate from entering active site
