Q

DNA sequencing steps

A

1) pcr to amplify sample, generate lots of dna fragments
2) add in regular deoxynucleotides give in the growing strands the substrate from which it can grow and dideoxynuclecitde to terminate elongation** , fluorescently label nucleotides with different labels, different sizes, different strands terminating at different points
3) electrophoresis to separate strands by size, computer analyses all the fluorescent

Q

PCR

A

Polymerase chain reaction, amplifying - makes many copies of particular fragment of DNA
- needed to clone into plasmid for example forensics and medical diagnosis
Goal is to make enough of the target of DNA can a analyses or used, gel electro, sequencing

Q

PCR Steps

A

1)Denaturation (95°C) - separates 2 strands, provides single stranded templates for next step
2) Annealing (55°C) - cool the reaction so primers can bond to their complementary sequences on the single stranded template DNA
3)
