Disease Progress and Drug Action Flashcards
What can clinical pharmacology be described as?
Understanding disease progress and drug action
What does drug action refer to?
- Time course of a drug (PKPD)
- link model to account for delays in relation to drug concentration
Whats a disease progress model?
Quantitative model that accounts for the time course of a disease status
Disease status = S(t)
How may disease status be defined?
Biomarkers
- Signs = physiological or biological measurements of disease activity
Clinical outcomes
- symptoms
- survival
What is the simplest disease model?
A linear model (natural history)
S(t) = So + A . t
What is the linear + offset (symptomatic) model?
A model that describes the delay in drug action effect and the washout of drug effect once the drug is stopped
Whats an example for the linear offset model?
ACh esterase inhibitors for alzhiemers
Whats the linear slope disease modifying model?
Accounts for when the disease causes damage / changes that are irreversible when medications are being used i.e the base level of disease changes
What are the disease models?
Linear
Linear + offset
Linear + disease modifying
