What is the reaction of detecting labelled enzyme antibody?
Ag + Ab → End Product Detection (LM)
Involves the use of only one antibody that is labeled. The labeled antibody is added to the antigen being detected. End product is observed under the Light Microscope.
Direct IHC
A more sensitive method. Modification of the direct/ traditional method. This involves addition of dextran polymer
Enhanced Polymer One-Step Staining Method (EPOS)
More sensitive than the direct method. A two-step method. Requires the use of 2 antibodies
Indirect IHC
Antibodies required in IHC: (2)
- Primary antibody
- Secondary antibody
This antibody used in Indirect IHC will attach to the Fc portion of the primary antibody, thereby increasing sensitivity
Secondary antibody
This antibody used in Indirect IHC is specific to antigen being detected
