D L2: Introduction to Chromatography Flashcards
when did chromatography start?
what is chromatography ?
A technique which allows the separation of a mixture of analyte molecules through their interaction with a stationary phase and a mobile phase.
what is the stationary phase?
The chromatographic phase (solid or liquid) that is held and through which the mobile phase moves
what is mobile phase?
The chromatographic phase that moves through the stationary phase, either a liquid or gas
what does eluent mean?
solvent entering a column or moving up a plate, becomes an Eluate when it leaves
whats elution?
process of passing a liquid/gas through a column
whats the analyse/solute?
the molecule undergoing separation Columns are either packed or open tubular
what is paper chromatogprahy used for ?
used to separate inks or food dyes
in paper chromatography what is the stationary phase and what is the mobile phase?
SP - paper
MP - water
Components in a mixture will move through the stationary phase at ____ _____ under the influence of a mobile phase.
different
rates
if a molecule acts with only the stationary phase, will there be any form of chromotogrpahy?
no there will not be because there needs to be two phases
if the analyte only reacts with the stationary phase where will it be ?
at the top
are both phases needed for chromtogrpahy?
yes
what is partition ?
the rate of movement of the analyte through the column is dependant upon the relative solubility of the analyte in the stationary phase and the mobile phase.
what is adsorption?
the relative polarities of the analyte and the stationary phase determines the rate at which the analyte moves.
