A
  1. place garlic clove in conc HCL for 5 mins in hot water bath (55 degrees)
  2. Take out and rinse thoroughly
  3. Cut several root tips off
  4. stain with acetic orcein in water bath for five mins to increase staining
  5. Add a drop of water and macerate to spread out cells
  6. count mitotic index

How to set up potometer

A

Fill capillary tube and rubber connector with water. UNDER WATER put stem of shoot into rubber connector. Smear petroleum jelly around the shoot and rubber to make sure airtight. Blott the leaves to make them dry.
