Method to observe mitosis in root tip
- place garlic clove in conc HCL for 5 mins in hot water bath (55 degrees)
- Take out and rinse thoroughly
- Cut several root tips off
- stain with acetic orcein in water bath for five mins to increase staining
- Add a drop of water and macerate to spread out cells
- count mitotic index
What does acetic orcein stain and do
Chromosones dark red and fixes the cells in place
why do we add filter paper to each Petri dish when investigating pollen tube growth
To act as a humid chamber so the pollen slides do not dry out

How to set up potometer
Fill capillary tube and rubber connector with water. UNDER WATER put stem of shoot into rubber connector. Smear petroleum jelly around the shoot and rubber to make sure airtight. Blott the leaves to make them dry.
How to set up respirometer
Soda lime to absorb CO2 , gauze and then organisms. Attach to tube and place coloured bubble at end
