Class #1 to #4 Flashcards
Effect of intra cellular receptors?
Alter gene transcription
Kd=
50% receptors bound
AVD
Vd=Dose / Drug [] in blood. High = more tissue binding. Low = more protein binding in blood
Efficacy=
How strong response after Dx bound to receptors
Eg of competitive partial agonist?
Oxymetazoline with Epi in nasal spray
therapeutic index=
LD50/ED50
Therapeutic window=
Concentration range where most people have therapeutic effect and very little have TD
Affinity vs Efficacy?
- Binding 2. USefullness
P Glycoprotein =
transport drug out of body
Metabolism two steps?
1)Hydrolysis/reduction/oxidation making compound less active or inactive. (Now lipid soluble)(Eg. AAS made into salicylic acid) 2) Conjugation of metabolite to make it more water soluble, transferases make rise to those conjugates (Eg. Gluthatione-s-transferase, glucoronic acid transferases).
P450 found where in hepatocytes?
ER
Family of CYP that metabolize drugs?
CYP1 CYP2 CYP3
Polymorphic distribution? Eg?
More than 1% of pop. has trait that cause differential expression of enz. metabolism.
Eg: CYP 2D6 - codeine, B-blockers, tricyclic antidepressants. CYP 2D6 inhibited by fluoexitine, haloperidol, paroxetine, quinidine
Excretion, mostly kidney?
Filtration (small molecules), secretion, reabsorption (lipid soluble are reabsorbed), water soluble are excreted.
Drug clearance?
Quantity eliminated in a given amount of time
