Sampling (selecting correct regions of the sample)
Polishing ( Uniform material removal = flat sample)
Etching (slective electro-chemical removal = non-uniform surface)

Why do some regions look bright and some dark in an optical micrograph?

A

In optical micrograph the main mechanism of contrast is reflection.
