What direction does Taq Polymerase
Taq Polymerase always copies from 5’ -> 3’ according to the “Nascient” Strand
(the newly formed strand)
Can Taq Polymerase bind to single stranded DNA?
NO! Polymerase can only bind to double stranded DNA, which is why primers are needed
What is the difference between Endonuclease and Exonuclease?
ENDONUCLEASE: cuts on dsDNA
EXONUCLEASE: cuts on ends of DNA

What is meant by bidirectional replication?
Both DNA strands are replicated simultaneously
(possible in both prokaryotic circular dsDNA and eukaryotic double helix dsDNA)

How does the spell check function work in DNA replication?
Polymerase detects an irregular base by its shape and stops then rocks back to allow exonuclease to come in and fix the mutation.
