Ch. 15 Confocal Microscopy Flashcards
What does the term “confocal microscopy through-focusing” refers to?
The CMTF intensity curve is then generated by calculating the average pixel intensity in a central region of each image, and plotting versus z depth. It then generates a curve that resembles that of an “A mode Ecogram” allowing for comparison in thickness and reflectivity of corneal layers. It also allows for the clinician to move the cursor over the curve, and se the corresponding en-face image to correlate.
Which are the two larger peaks in a normal CMTF curve
normal CMTF (confocal Microscopy through-focus) have two main peaks representing epithelium and endothelium. A
What findings can be seen in a report of a confocal microscopy in a patient with acanthamoeba infection?
Two types of hipereflective structures (trophozoites and cysts)
can bacteial and viral infections be imaged usind confocal microscopy?
No.
The current main indications in infectious keratitis are Acanthamoeba and fungal keratitis.
What is a Second Generated Harmonic Signal?
To generate an SHG signal, a material is excited by the simultaneous absorption of two photons of light of the same frequency and then the material emits a single photon of light that is double the frequency or half the wavelength of the excitation light. The ability to generate SHG signals is limited to materials that are highly ordered and noncentrosymimetric.
How can SHG has been used to study the cornea?
Second generated Harmonic signal has been used to study the collagen structure of the cornea.
What differences can be found in Second Harmonic Generated Harmonic of keratoconic corneas?
Less collagen interweaving.
Less fibers attached to the anterior limiting lamellae
Less fibers attached to the posterior stroma (sutural fibers are decreased)
Overall number of fibers is decreased in the anterior stroma.
