Autoimmune Diseases Flashcards
Is autoimmunity genetically determined?
Yes
What causes autoimmune diseases?
Breakdown of self-tolerance
Where is T cell auto-immunity removed?
Thymus
What may dispose a T cell to autoimmunity?
Class 2 MHC genes
What are other causative associations?
> Sex (Women > Men)
Older age
Sequestered (hidden) antigens - eye, testis
Environmental triggers- smoke, infection, trauma.
What two responses can infections have?
Autoimmunity and normal immune response.
What is molecular mimicry?
When similar sequences between foreign and self-peptides are enough to activate auto-reactive B/T cells
An example of molecular mimicry includes?
RHEUMATIC FEVER
- Antib’s against streptococcus damage heart glycoproteins.
What can changes in the nature/number of autoantigens cause?
Autoimmunity
What makes proteins more immunogenic?
Citrullination
What binds to HLA-DQ to cause Coeliac Disease?
Tissue transglutamase altered gluten
What does failure to clear apoptotic debris do and cause ?
Increase availability of antigens inside the cell. Causes Systemic Lupus Erythematous.
How do T cells cause inflammation?
Cytokines.
Helping B cells make autoantibodies.
What do autoreactive B cells and their antibodies activate?
Activate complement cascade
What do autoreactive T cells do?
Release cytokines
