Antifungals Flashcards
Azole drugs?
Fluconazole
Voriconazole
Posiconazole
etc..
Echinocandin drugs?
Caspofungin
Micafungin
Aniladulafungin
Polenes drugs?
Amph B
Nystatin
MOA of azoles?
inhibits cytochrome lanosterol P450 14 alpha demethylase which prevents ergosterol synthesis (an essential component of cell membrane)
MOA echinocandins?
Inhibits beta 1,3 D glucan synthase (an essential component of cell wall) which prevents cell wall synthesis
MOA polyenes?
2 MOA
- Disrupts cell membrane by binding to ergosterol which disrupts activity in cell membrane causing leaking of cell content - cell death
- Oxidative damage to fungal cells (causes oxidative burst - damages fungal cell)
MOA fluorouracil (5FU) - flucytosine?
prevents DNA and protein synthesis
MOA terbinafine?
inhibits squalene epoxidation preventing formation of ergosterol
MOA of ibrexafungerp?
Similar to echinocandins but binding site are different
MOA fosmanogepix?
inhibits fungal Gwt1 protein - prevents GPI anchor biosynthesis pathway - affects cell wall integrity
MOA griseofulvin
disrupts mitosis by acting on microtubules
MOA pentamidine
interacts with DNA, RNA, topoisomerase and ubiquitin
Fungus with amphotericin resistance?
Candida lusitinae
Candida guilliermondii
Aspergillus tereus
Scedosporium prolificans
Trichosporon asahii
