Aminoglycosides Flashcards
What are the drugs under the Aminoglycoside drug class?
Streptomycin
Gentamycin
Tobramycin
Neomycin
Amikiacin
What is the MOA of Aminoglycosides?
Binds to the 30s ribosomal unit which disrupts the assembly of the ribosome, causing it to not function properly or misread the genetic code (mRNA) which makes this class of drugs bactericidal
What is the SOA for Aminoglycosides?
Aerobic Gram (-) bacteria such as Pseudomonas, Klebsiella, and Enterobacter species
Can treat Gram (+) bacteria in infections such as Infective Endocarditis when used in combination with beta-lactams or Vancomycin
What are the adverse effects of Aminoglycosides?
Ototoxicity
Nephrotoxicity
How do Aminoglycosides enter the bacteria?
Through porin channels in the outer membrane of bacteria. Once inside, they need an oxygen-dependent transport system to cross into the inner membrane. Aerobic bacteria have these transports whereas anaerobic bacteria do not which will make the bacteria resistant to aminoglycosides.
What does it mean to be concentration dependent killers?
Aminoglycosides are concentration-dependent killers, meaning their effectiveness depends on how high the drug concentration is.
