Benzodiazepines
Bind to a subunit of GABA-A receptors and facilitate GABA transmission (increase likehood of Cl- channel opening). Many many forms but 3 mains groups :
Barbiturates
Bind to physically different site on the receptor than benzodiazepines and facilitate GABA transmission (increase likehood of Cl- channel opening and duration of opening). Ex : pentobarbital
test anxiety ?
induce social fear, avoidance conflict test, latency to go to food pellet
Antidepressants ?
trycyclic, SSRI : serotonine selective reuptake inhi. Citalopram, fluoxetine, fluvoxamine, paroxetine, sertraline
NRI : reboxetine
SNRI (mixed 5-HT / NE) : Serotonine + norad transporters. venlafaxine, duloxetine, milnacipran
MAOI : tranylcypromine, phenelzine
Others : bupropion, mirtazepine, trazodone, ketamine (NMDA antagonist)
ECT (electro convulsivo therapy): 6-8 generalized seizures over 2-3 weeks
Transcranial magnetic stimulation (TMS)
Transcranial direct current stimulation (tDCS) : not invasive, just on the brain
tricyclic
amitriptyline, imipramine, clomipramine (> 5-HT)
nortriptyline, desipramine (> NE)
Serotonine + norad transporters
type of validity to look for ?
Faith validity : same symptomes
Construct/Pharmacological validity : drug act on the same way in model and human
Aetiologic/Predictive validity : same biological marker
Learned Helplessness model
Alteration du REM sleep, weight loss, decreased sexual behavior, Increase levels of CRF and corticosterone. Sensitive to antidepressants and ECT.
