What does tube IAT consist of

A

-detects antibodies (cold and Warm)
-Read at IS, 37, and AHG
-Crossmatching (Full IAT) Prewarm Technique IAT if cold antibody is present

A

Positive Antibody Screen
must have

A

-ID all AB detected regardless of significance because one AB can mask the other.
-serum/plasma tested against PANEL CELLS (unknown with known phenotype)
-must have an auto control cell where we test the pt cells with their own plasma

What should panel cells have

many positive cells, few, variation, auto control negative, auto control positive

A

minimum of 10 cells (so you can ID a pattern of homo/hetero cells of all significant AB)

many positive- multiple AB or AB to high incidence AG

few positive - AB to low incidence AG or weak AB showing dosage

Variation in strength- AB showing dosage

Autocontrol is neg- Allo AB

Auto control is pos - Allo AB or Pt AB has been sensitized

A

A
  • look at non reacting cells
    β€œreverse elimination logic”
    -if strong AG (homo expression) did not react with pt plasma then the AB to that AG is not present
    -0 reactions are negative

A

if hetero JKa JK (a+b+) did not react then Anti Jka or Anti Jkb are less likely to be present in pt plasma

-since it can show dosage it is not eliminated and listed under least likely of AB
