trangenic plants (tDNA) Flashcards
what is a common bacteria used to make transgenic plants?
agrobacterium tumefacians as it naturally infects DICOTS and creates a crown gall
rhibozium is better at infecting monocots
decribe the TI plasmid
it has left and right border tandem repeats and 3 genes auxin, cytokinin and opine which help to signal replication
we can use the left and right bordrs in biotechnology and virulence region to facilitate movement of tdna into the plant genome
what are the requitemetns of a vector
need an origin of replication and a promoter sequence to ensure the gene is transcribed)(expression vector)
Unique cutting site
need a selectable marker to show that the plasmid has been integrated into genome and select for the transformed plant
what is the difference between stable and transient transformation?
stable - growing plant from seed to soil, quite slow but you can isolate pure transformed strains
transient - you inject a growing plant with a vector but this is not permanently inherited
why are helper plasmids used in transformatin?
a plasmid with both a virulence region and the target gene is very large so we can use 2 plasmid vectors, the main one and the helper one with the vir region
Main one with gene insert, left and right borders and the MCS
We DON’T NEED THE TDNA
