topic 1 - enzyme controlled reactions Flashcards
cgp (topic 1A) 14 - 15
what is the first step to measuring the rate of an enzyme controlled reaction
set up boiling tubes containing the same volume and concentration of hydrogen peroxide - to keep pH constant add equal volumes of a suitable buffer solution to each tube
what is the second step to measuring the rate of an enzyme controlled reaction
set up the rest of the apparatus - a trough of water with an upside down measuring cylinder inside - a boiling tube containing hydrogen peroxide solution and catalase enzyme with a bung at the opening and a delivery tube attached to that, which has been also put through the upside down measuring cylinder
what is the third step to measuring the rate of an enzyme controlled reaction
put each boiling tube in a water bath set to a different temperature along with another tube containing catalase - wait 5 minutes before moving onto the next step so the enzyme get up to temperature
what is the fourth step to measuring the rate of an enzyme controlled reaction
use a pipette to add the same volume and concentration of catalase to each boiling tube - then quickly attach the bung and delivery tube
what is the fifth step to measuring the rate of an enzyme controlled reaction
record how much oxygen is produces in the first minute of the reaction - use a stopwatch to measure the time
what is the sixth step to measuring the rate of an enzyme controlled reaction
repeat the experiment at each temperature three times and use the results to find an average volume of oxygen produced
what is the seventh step to measuring the rate of an enzyme controlled reaction
calculate the average rate of reaction at each temperature by dividing the volumes produced by the time taken
