To prepare and examine plant cells, using a light microscope Flashcards
When preparing and examining a plant cell, what plant did you use?
onion
What’s your first step of preparing?
cut onion and remove the outer dry/scaly leaves
Using your finger/forceps, what do you do after cutting your onion?
pull a strip of thin, transparent epidermis from the inner curve of a fleshy, inner leaf
Where do you put the epidermal strip?
on a glass slide
Add a few drops of what to the slide with the epidermis? What colour is this? What colour does epidermis do?
iodine solution
red-yellow
nucleus goes orange, cytoplasm goes yellow
What do you add over the glass slide?
add a cover slip
How do you add the cover slip? Why this way?
at an angle to eliminate air bubbles
2 uses of adding a cover slip?
prevent the cells from drying out
prevents the lens from getting stained
When examining the slide, where do you put it and how is it secured?
place on stage, securing with clips
What objective lens do you use first?
lowest power
How do you focus on the image?
using coarse focus wheel
How do you finely focus on the image?
focus wheel
What are the results of looking at the cells under the microscope?
nucleus is dark brown
