Tissue culture generally works on the basis that a sample of a tissue is removed from a plant or animal.

Cells are grown in a bath of sterile fluid containing a source of nutrition
Why must the environment for the cells to grow be sterile? Why is this necessary?
This is necessary as micro-organisms might grow faster than the desired cells and they might contaminate the product.

Tissue culture is the growth of cells in or on a sterile nutrient medium outside an organism
