16
Q

What is the light source?

A

A microscope must have a source of light. Some light sources are built in the microscope itself

17
Q

What is the stage?

A

Area where the slide is viewed from

18
Q

Stage clips are used for?

A

Holding down the slide

19
Q

What is a diaphragm?

A

Adjusts the amount of light

20
Q

Course adjustment screw is used for?

A

Focusing on specimen when SMALLEST OBJECTIVE LENS IS USED

21
Q

Fine adjustment screw is used for?

A

Focusing on the specimen when the other lenses are used

22
Q

How do you use a microscope?

A

STEP 1: Turn on the light

STEP 2: Make sure the lowest lens is in place. Place slide on centre of the stage

STEP 3: Clip the slide in place

STEP 4: Look through optic tube with coarse screw to get the best view of specimen

STEP 5: Use fine screw to get a sharper image

STEP 6: When viewing at higher magnification, use x10 lens to view with fine adjustment screw.

23
Q

How do you prepare a wet mount?

A
  1. Make sure slide and coverslip are clean
  2. Place large drop of water in centre of slide
  3. Put specimen over droplet
  4. Carefully at a 45* angle place the cover slip over specimen
  5. Try not to get any air bubbles, if there are air bubbles you may have used too much water or specimen is too thick or it was folded.
  6. Wipe any excess water away and make sure the slide is dry
24
Q

List the characteristics of a good slide

A
Clean and dry
No air bubbles
Specimen not folded
Specimen is in the centre of the slide
Specimen is not too large or thick
25
Q

What are the possibilities for air bubbles?

A

Specimen is too big
Specimen is folded
Too little water used
