Agglutination test
A
Check for specific antibodies in a serum sample
Serum exposed to antigen-bound latex beads/RBCs in a range of dilutions

Complement fixation test
A
Used to investigate serum for antibodies against a specific antigen
The antibodies ‘use up’ the added compliment when the antigen is added
So when RBCs (prebound to antibody) added, there is no complement to lyse them
A series of complement dilutions are tested
A serum with plenty of antibody will avoid RBC lysis at a high conc vs serum without the antibody

How to get pfu/ml from plaque assay
A
count colonies
times by dilution (eg if 1x10⁻⁷, then times by 1x10⁷)
That is in given volume, convert to per ml
