What should you label an agar plate with?
The date, your initials, the name of the culture (e.g. sample A)
How do you ‘streak out’ a bacteria colony?

Flame a wire loop
Using the loop pick up a single colony and then streak it on to the fresh agar plate by using a back and forward movement
First draw two lines then streak out for a couple lines
Turn the agar plate and repeat the process making sure you overlap each side
What should you do after streaking out your colony?
You need to incubate your plate at whatever temperature is required (usually at 37 degrees)
Why is streak-inoculation done ?
It is done to achieve a pure culture which can be subjected to basic characterisation and further tests
