Special Stains (Carbohydrates) Flashcards
Stains Mucin
Best Carmine Method
Mucicarmine Stain
Differentiates Acid Mucin from Neutral Mucin
Combined Alcian Blue-pas-hematoxylin Technique
Stains Sulfated Mucins
Gamori’s Aldehyde
Stain Acid Mucin
Colloidal Iron
Fluorescent Arcidine Orange
Stains Encapsulated: C. Neoformans
Southgate’s Mucicarmine Technique
Stains Glycoaminoglycans
Metachromatic Staining - Azure A
Metachromatic Toluindine Blue
Best Carmine stains the following
Glycogen
Mast Cell Granules
FIBRIN
MUCIN
Used for the demonstration of GLYCOGEN specially when it uses the Diastase Method; may also stains MUCOPROTEINS, MUCIN and PHOSPHOLIPIDS
Periodic Acid Schiff (PAS)
Has a good affinity to glycogen (bright red) due to alkaline carminic acid
BUT weakly stains mast cell granules, fibrin, mucin
Best Carmine Method
An OBSOLETE STAIN and NOT SPECIFIC for glycogen.
Rapid but tends to fade after few months
Langhan’s Iodine Method
Stains Glycoaminoglycans and gives RED-PURPLE stains
Metachromatic Staining - Azure A
Metachromatic Toluidine Blue
Most popular method for ACID MUCIN (+ blue )
Alcian BLUE Technique
Differentiates Neutral Mucin from Acid Mucins
Combined Alcian Blue-PAS-Hematoxylin technique
PAS = Neutral Mucin
Alcian Blue = Acid Mucin
Demonstrate mucin by Adding ALUMINUM HYDROXIDE to Carmine
Mucicarmine stain
This stain carbohydrates and absorbed onto tissue containing ACID MUCIN at LOW pH and visualized by its conversation to FERRIC CYANIDE
Colloidal (Dialyzed) Iron Technique
