Compare bulk and single cell sequencing.

A

Bulk:
- No heterogeneity but higher throughput
- No VDJ-VJ pairing (exist on diff. chromosomes)
- No clonal expansion (one cell can produce more mRNA than another)
- Error-prone (during PCR and seq.)

Single Cell:
- Heterogeneity, lower throughput, higher resolution per cell/clone
- VDJ-VJ pairing via: droplet-based microfluidics, primer-matrix barcoding, emulsion PCR, combinatorial demultiplexing, and 10xGenomics droplet seq.
- Clonal expansion possible
- Linking genotype (BCR/TCR sequence) to phenotype (binding or not)
- Prevent errors via UMIs
