What are bnAbs?
A
Broadly Neutralizing Abs
- Show a high rate of SHM compared to other BCR/TCR CDRs
- Are polyreactive (able to bind a variety of antigens with low affinity)
- Used to develop HIV vaccine
- Isolate them from elite neutralizers, expose them to HIV antigen
- Can also sequence mature bnAbs and work backwards to pre-SHM state, target this form + design immunogen to select for desired bnAb lineage

1) Hybridoma + ELISA immunoassay
2) Single Cell Sorting by FACS w/GFP or intracellular staining
