Is a process whereby tissues are cut into uniformly thin slices or “sections”
SECTIONING
The excess wax is cut off from the block to expose the tissue surface in preparation for actual cutting
TRIMMING
Paraffin embedded tissue blocks cut by rocking and rotary microtomes.
Sections are cut between 4-6 microns in thickness.
PARAFFIN SECTION
Also called collodion or parloidin, it is a nitrocellulose.
CELLOIDIN SECTION
Celloidin embedded tissues cut by sliding microtome
CELLOIDIN SECTION
May be cut from tissues that have been fixed and frozen with CO₂ or for fresh or fixed tissues frozen with the cryostat to provide immediate report of the tissue sample

To do enzyme immunocytochemistry and immunoflourescent studies

Point at which sections may then be cut at 10 micra thickness

Most common method of freezing
Adequate for freezing small pieces of tissues except muscle.
AEROSOL SPRAYS
Floatation bath temperature
-6°C-10°C lower than the melting point of the wax
Required on sections that are to be exposed to acids and alkalis, thus giving a “dirty” background to sections
