Dehydrating agents Flashcards
especially used for tissue sections thatgmentary biopsies require urgent examinations such as fra
A very concentrated solution (above 80%) makes tissues
Hard,brittle and difficult to cut
ALCOHOL
Routine; fast; cheap
Clear,colorless,flammable fluid
Ethanol
Is a toxic dehydrating agent
For blood (does not lyze RBCs) and tissue films and for smear preparation
Methanol (methyl alcohol)
Slow dehydrating agent
For plant and animal microtechniques
Less shrinkage and hardening effect
Recommended for tissues which do not require rapid processing
Butanol (butyl alcohol)
Substitute for alcohol
Very rapid (urgent biopsies)
Acetone
Dehydrates rapidly
Ethylene glycol ethers are combustible at 110-120°F and are toxic by inhalation, skin contact and ingestion
Cellosolve (ethylene glycol monoethyl ether)
Fast
Does not harden tissue
Produces very little distortion
Minimal shrinkage
Soluble in alcohol, water ,ether ,benzene ,chloroform ,acetone, and xylene
Triethyl phosphate
Acts both as a dehydrant and clearant
Produces less shrinkage than alcohol
Expensive
Toxic to the liver
Dioxane (diethylene dioxide)
Has a very offensive odor
Vapor causes nausea,dizziness, anesthesia
Tetrahydrofuran
The process of removing intercellular and extracellular water from the tissue
Done following fixation and prior to wax impregnation
For routine tissues and if infiltration is with non-water miscible medium.
