RP6-aseptic techniques Flashcards
state 6 aseptic techniques
(WBFFCK)
-wipe down all surfaces before and after experiment with antibacterial cleaner
-use a Bunsen burner in work space so convection current draws microbes away from culture (inoculating loop)
-flame the neck of the McCartney bottle to prevent bacteria entering vessel
-flame hoop before using it to transfer bacteria
-close windows to prevent air currents
-keep all vessels containing bacteria open for a minimum amount of time possible
why is the bacteria cultured at 25C
-prevent growth of harmful bacteria that occur at higher temps
How can you compare the effectiveness of different antibiotics applied to the same bacteria
-measure the diameter and calculate the area of the zone of inhibition on the agar
hazards and precautions of the aseptic techniques practical
-naked flame, burns, handle with care or run under cold water for 20mins
-bacteria, biohazard, use disinfectant and wash hands
-disinfectant, flammable, keep away from naked flames
why should the lid not be completely taped to the petri dish
-allows oxygen to enter the petri dish and prevents growth of harmful anaerobic bacteria
