reticulin stain Flashcards
1
Q
what collagen type makes up reticulin
A
type III collagen
1
Q
what is reticulin
A
supporting mesh or organs and tissues, particularly liver, lymphatic system, bone marrow
2
Q
reticulin is a sensitive indicator of
A
organ damage
- staining is used extensively for liver pathology
3
Q
what is the principle of reticulin silver staining
A
- metallic impregnation
- silver reacts with active groups on reticulin
4
Q
steps of reticulin staining
A
- Oxidation (Potassium Permanganate)
- Bleaching (Oxalic Acid)
- Sensitization (Ferric Ammonium Sulphate)
- Silver Impregnation (ammoniacal silver)
- Reducing (Formalin)
- Toning (Gold Chloride)
- Rinsing (Sodium Thiosulphate)
- Counterstaining (Eosin)
5
Q
what are some precautions of reticulin staining
A
- amoniacal silver is preapres fresh and very sensitive to pH
- sensitization agen is tenuous and should not be subject to aggressive washing
- unreduced silver left on slide will darken with time
- high basic solutions may cause tissue to lift off slide
