Restriction enzymes that break or restrict DNA polymer at the sugar phosphate backbone
Endonuclease
My lease and methylated activity, binds to host specific DNA, 4-6bp sites containing methylated adenines
Type 1 restriction endonuclease
Methylated and restrict (cut) DNA recognition sites are asymmetrical, cleaves at 24-26bp from the site to the 3’ end
Type 3 restriction endonuclease
Cutting and methyltransferease function
Type 4 restriction endonuclease
Most used in the lab, no inherent methylation activity, bind to simple diners of symmetrical DNA recognition site (palindromic), cleave DNA directly at binding site
Type 2 restriction endonuclease
Metabolizing enzyme that degrades DNA from free 3’ hydroxyl or 5’ phosphate ends, not circular DNA, make stepwise deletions to modify specific ends after restriction enzymes cut
Exonuclease
Metabolizing enzyme that degrades single stranded DNA FROM 3’ hydroxyl end into mono nucleotides, optimal for long ends
Exonuclease 1
Metabolizing enzyme that removes 5 mononucleotides from 3’ ends of double stranded DNA, needs the presence of Mg2+ and Mn2+, has some endonuclease activity, removes from blunt ends or nicks but not overhangs
Exonuclease 3
Metabolizing enzyme that digests single stranded DNA from 5 or 3 end to remove long protruding strands
Exonuclease 7
Enzyme that catalyzes the formation of phosphodiester bonds
DNA ligase
