What are restriction enzymes?

A

Each restriction enzyme will only cut DNA strands at recognition sites - specific bases sequences

  • When DNA is cut with a restriction enzyme the resulting fragements are left with either a short overhang of a single DNA called STICKY ENDS
  • Or no overhanging DNA which is called a BLUNT END

A
  • Ligase joins DNA or RNA together by creating bonds in the sugar-phosphate backbone
  • Ligase is a group of enzymes that work with restriction enzymes to join together DNA, hydrogen bonds form between complementary bases of sticky ends
