Restriction digest Flashcards
1
Q
Restriction digest
A
restriction endonucleases (aka restriction enzymes)
2
Q
What are restriction enzymes?
A
Each restriction enzyme will only cut DNA strands at recognition sites - specific bases sequences
3
Q
Cutting specifically
A
- When DNA is cut with a restriction enzyme the resulting fragements are left with either a short overhang of a single DNA called STICKY ENDS
- Or no overhanging DNA which is called a BLUNT END
4
Q
Ligase
A
- Ligase joins DNA or RNA together by creating bonds in the sugar-phosphate backbone
- Ligase is a group of enzymes that work with restriction enzymes to join together DNA, hydrogen bonds form between complementary bases of sticky ends
5
Q
Ligase with sticky ends
A
stick ends are sealed together but gaps remain
6
Q
Ligase with blunt ends
A
Ligase seal the gaps in blunt ends
