Recombinant Plasmids Flashcards
Recombinant plasmids function
For a gene to be expressed it needs to be inserted into a living cell - such as a bacteria
- A plasmid ( small piece of circular DNA that is naturally found in bacteria) can be used as a vector to insert a gene such as the human insulin gene into bacteria
Vector
an agent or vehicle used to transfer pathogens or genes between cells and organisms
PROCESS OF CREATING A RECOMBINANT
1) The DNA is extracted a from cell, cut with restriction enzymes to create sticky ends
stage 2)
The DNA attaches to the plasmid as there complementary bases.
3)
Ligase is added to the fragments to make the joining permanent
4)
plasmids that incorporate the foreign DNA = have become recombinant plasmids.
some just wont form recombinant plasmids
5)
Bacterial cells are treated to make their cell walls permeable to plasmids so that they can enter the bacteria
6)
Bacteria cells with recombinant plasmids = transformed bacteria
