Quinolones Flashcards
Source and chemistry
synthetic.
quinolones are fluorinated except Nalidixic acid and Cinoxacin
Absorption
well absorbed except in the presence of antacids containing Al3+ and Mg2+.
Distribution
Poor passage across BBB and Pass placental barrier.
Metabolism
liver EXCEPT Ofloxacin.
Excretion:
- Urine
2.biliary
Action on bacteria
Bactericidal.
Mechanism of action
They inhibit DNA synthesis by inhibition of DNA gyrase (Topoisomerase II) which is responsible for supercoiling of bacterial DNA.
1st generation Quinolones:
Nalidixic acid).
2nd generation Quinolone
Cinoxacin
3rd generation Quinolones
Fluoroquinolones):
1. Norfloxacin
2. Ciprofloxacin
3. Ofloxacin
New fluoroquinolones
- Levofloxacin
- Trovafloxacin
- Cinfloxacin
Adverse effects:
1.Reversible arthropathy (damage of growing articular cartilage of bones), tendonitis, and joint swelling.
