If the wells are round that is good as we can see a shape of the sediment in the bottom if it is positive or negative. if they are flat all the sediment will go to the bottom and we cannot determine a negative or positive result.

Serum is the liquid that remains after the blood has clotted.
Plasma is the liquid that remains when clotting is prevented with the addition of an anticoagulant.
