Proteins as Drug Targets Ch 2 Flashcards
Primary structure
Order of amino acids
Peptide bond property
Planar amide bond
Secondary structure
Local folding (alpha helix, beta sheet, and beta turns)
Tertiary structure
Overall 3-D shape of protein as a result of hydrophobic forces and covalant bonds (disulfide bridges).
Quaternary structure
How individual polypeptide chains (subunits) associate.
Post-translational modification
Phosphorylation, glycosylation, and amino acid modification that occurs after the biosynthesis of protein.
Translation
Process by which protein is biosynthesized
Proteonomics
Identification of all the proteins in the cell
Monoclonal antibodies (2 examples)
Can be chimeric (from mice) or fully humanized. They are not proteolyzed quickly in the body. Ex. Herceptin, Avastin.
