Pre-Lab #7 Flashcards
- LB agar plate inoculated with Escherichia coli -pGLO.
a. Do you expect to see bacteria? Explain why or why not.
b. If you expect to see bacteria, will they glow? Explain why or why not.
i. Yes you would expect to see growth because the LB agar gives the nutrient rich environment and
i. If they are expressing GFP then they will glow when in the presece of UV light
- LB agar plate containing ampicillin inoculated with Escherichia coli -pGLO.
a. Do you expect to see bacteria? Explain why or why not.
b. If you expect to see bacteria, will they glow? Explain why or why not.
i. No you wouldn’t expect to see bacteria. Since pGLO gives the gene resistance to ampicillin, with there being no pGLO there would not be the risastance to the ampicllin so it woud not be a good growth area for the the bacteria
i. No growth so no glow
- LB agar plate containing ampicillin inoculated with Escherichia coli +pGLO.
a. Do you expect to see bacteria? Explain why or why not.
b. If you expect to see bacteria, will they glow? Explain why or why not.
i. Yes you would expect to see bacteria, because the pGLO is present so they can grow in th eampicillin plate since they are amipcillin resisant
i. No it shouldn’t glow because it only has ampicillin, not both that and arabinose.
- LB agar plate containing ampicillin and arabinose inoculated with Escherichia coli +pGLO.
a. Do you expect to see bacteria? Explain why or why not.
b. If you expect to see bacteria, will they glow? Explain why or why not.
i. Yes you would expect to see growth because the pGLO plasmid contains a gene that
i. Yes it should glow if UV light is shined. Since the plate contains ampicillin and arabinose the bacterial growth should glow.
