Mechanism of action
Blocks voltage - dependent sodium channel as well as AMPA and glutamate Rc.
Is a weak carbonic anhydride inhibitor
TOPIRAMATE and ZONISAMIDE increase the risk of renal stone formation.
ZONISAMIDE
Is a sulfa anticonvulsivant that blocks sodium and calcium channels

NPC1L1 (Nieman Pick C1 like peptide)
Expressed at the gut lumen (apical) and the hepatobiliary ( canalicular ) interface in the intestinal epithelial cells.
This structure facilitates the internalization of free cholesterol into the enterocyte in conjunction with the adaptor protein 2 (AP2) complex.
EZETIMIBE
Inh NPC1L1 and decreases the delivery of intestinal cholesterol to the liver and reduces hepatic cholesterol stores resulting in increase in clearance of cholesterol from the blood
Also significantly lowers LDL level and is often combines with statins (HMG -CoA REDUCTASE INH) to achieve LDL TARGET.
EZETIMIBE
Adverse effects
Upper respiratory tract infections
Sinusitis
Has no clear effect on mortality
NPC1L1 also found in the upper respiratory tract

When ZOLENDRONATE is indicated
Is parentally administered bisphosphonate that is indicated in patients who are intolerant to oral therapy.
Or in whom orally administered are inappropriate such as those with Celiac disease, achalasia, or inflammatory bowel disease.
