PEPCK regulation Flashcards
PEPCK important regulation why?
can produce something that can be shared among other pathways
PEPCK control
- transcriptional control, no longer allosteric:
Insulin inhibit PEPCK gene transript via PI3K signaling cascade (insulin pathway)
cAMP made in response to AC (2nd mess to glucagon.. low BS) which induces/activate PEPCK gene transcription
How are PEPCK and PKL similar in control?
Eukaryotic PEPCK isoforms
2 isoforms. Cytosolic PEPCK1 is important because it is the one that is hormonally regulated via insulin and glucagon.
Covalent modification PEPCK
Can add an cetyl group to a Lys in PEPCK1, which is supposed to cause PEPCK1 to be degraded (ubiquitin), but in this scenario it DOES NOT.
INSTEAD:
bc of High glucose causing this, will enable PEP to be converted backward to OA. Like PC can be an anapleurotic…
This will happen when high Glucose quantity becauyse gluc will be converted to pyr which is converted to Ace-CoA then enter TCA which needs OA to enter.
