PCR Cycle Flashcards
1
Q
General description of the three stages
A
- Desaturation of DNA template
(~94C to 95C) - Annealing (~50C to 60C)
- Extension/ elongation (~72C)
2
Q
Describe denaturation
A
- H bonds break;
- target DNA template denatures into ss;
3
Q
Describe annealing
A
- excess DNA oligonucleotide primers anneal to DNA at opposite ends of target seq;
- complementary base pairing
4
Q
Describe extension
A
- taq attaches and adds dNTPs to 3’end of primer;
- new strands synthesised in 5’-> 3’ direction; complementary to the template
5
Q
Why PCR is a chain rn?
A
Newly synthesised DNA strands serve as templates for further DNA synthesis
6
Q
What if the length of the primers are too short?
A
Hybridise to non-target sites
-> undesired amplification pdts
