Parkinson's Flashcards

1
Q

How is iron involved in PD?

A

May accelerate ND via OS.
OR increased OS may release iron from its stores.
Expose neonatal mice to iron –> Parkinson-like nd with age.

Q

What is Parkin and how is it involved?

A

An E3 ubiquitin ligase - Norris et al (2015)
Interacts with PINK1 to promote mitochondrial fission.
Under moderate stress stimulates connectivity.
Under high stress triggers mitophagy.

Q

What is PINK-1?

A
A ser/thr kinase - causes Parkin to bind depolarised mt - induces mitophagy

Q

What is DJ-1?

A

OS sensor
relocates to mt under OS conditions and protective.
Mutants –> increased sensitivity to rotenone, paraquat and H2O2.

Q

What did Zhang show?

A

High VE intake –> reduced risk of PD (2 large cohorts = nurse’s health study, health professionals follow up).

Q

What about Shults et al.?

A

8- patients with early PD –> 16 mths intervention with CoQ –> slow progressive deterioration of function in PD.
