Characteristics of Beckwith-Wiedemann syndrome (6)
- Growth disorder (neonatal hypoglycemia, macrosomia, macroglossia, hemihyperplasia)
- Omphalocele
- Embryonal tumors (Wilms tumor, hepatoblastoma, neuroblastoma, rhabdomyosarcoma)
- Visceromegaly, adrenocortical cytomegaly
- Renal abnormalities (medullary dysplasia, nephrocalcinosis, medullary sponge kidney, and nephromegaly)
- Ear creases/pits

Cardinal symptoms of Sotos syndrome? (3)
- Facial appearance: broad and prominent forehead with a dolichocephalic head shape, sparse frontotemporal hair, downslanting palpebral fissures, malar flushing, long and narrow face, long chin
- Learning disability: early developmental delay, mild-to-severe intellectual impairment
- Overgrowth: height and/or head circumference ≥2 SD above the mean):born LGA and rapid growth for initial 5 years

Diagnosis?
- Neuro: Cranial MRI/CT abnormalities, seizures, behavioral problems (most notably autistic spectrum disorder)
- Cardiac anomalies
- Renal anomalies
- MSK: Joint hyperlaxity, advanced bone age, pes planus, scoliosis
- Maternal preeclampsia, neonatal complications

- Loss of methylation on the maternal chromosome at imprinting center 2 (IC2) in 50% of affected individuals
- Paternal uniparental disomy for chromosome 11p15 in 20%

Diagnosis?
- Progressive segmental overgrowth (the skeleton, skin, adipose, and central nervous systems), progresses rapidly in the toddler, severe disfigurement
- Tumors (lipomas, lymphagiomata, hemangiomata)
- Pulmonary complications
- Deep vein thrombosis and pulmonary embolism
