- does not accumulate
Tacrine
- non-selective inhibitor of both AChE and BChE
- non-competitive
Amantadine
- used in PD
- augments dopamine neurotransmission- not clear how
Dextroamphetamine and methylphenidate
- indirectly induce release of dopamine and noradrenaline from presynaptic neurons
- also weak inhibitor of catecholamine reuptake
- inhibitors of MAO
Atomoxetine
-selective inhibitor noradrenaline reuptic (NARI) similar t reboxetine
Carbidopa
- inhibits aromatic-L-amino acid decarboxylase (DOPA decarboxylase)
- cannot cross the BBB so is given with l-dopa
Dantrolene
- directly affects the formation of actin-myosin complexes in skeletal muscle through ryanodine calcium channel inhibition
- muscle relaxer
Diphenydramine, promethazine
-H1 receptor antagonist
Levodopa
-combined with carbidopa to reduce peripheral conversion to dopamine
modafinil
-activates hypocretin-producing neurons (alerting effects)
Pemoline
indirectly stimulates dopaminergic activity
- stimlant
- not used due to hepatotoxicity
Reserpine
- depletes stored dopamine and other monoamines
- can lead to depression and suicide
- antihypertensive
