Nucleases
Enzymes that cut DNA by breaking phosphoester bonds linking nucleotides
Exonucleases
Nucleases that remove nucleotides one by one from the end of the DNA strand
Exonuclease III
Degrades DNA from 3’ to 5’. Can create single strand DNA
Endonucleases
Break internal bonds within DNA
S1 nucleases
Only cleaves single strands
DNase1
Cuts both single and double strands, can be used to locate sections protected by other proteins
Restriction enzymes
Endonucleases that cut only after a specific base sequence
Modifying enzymes
Add or remove specific groups
Alkaline phosphatase
Remove phosphate from 5’ end of chain
Polynucleotide kinase
Transfers phosphate from ATP to the 5’ end
Terminal deoxy-nucleotidyl transferase
Adds nucleotides to the 3’ end of DNA strands
Klenow fragments
Can remove 3’ overhangs and fill in 5’ ones.
