Non-Oncogene Addiction Flashcards
1
Q
Give examples of non-oncogene addictions in cancer cells
A
Dependence on heterotypic signalling Dependence on prosurvival pathways Dependence on angiogenesis Glucose dependence Dependence on stress supports; oxidative stress, dna damage stress, mitotic stress, proteotoxic stress
2
Q
What is velcade/bortezomib?
A
Proteosome blocker
Main target is chymotrypsin-like activity at the beta5 subunit (PSMB5)
3
Q
How does velcade/bortezumab work?
A
Prevents proteosome activity and so destruction of dysfunctional proteins and so disrupts the protein homeostasis of the cell, submitting it to proteosome stress.
4
Q
Which cancers does velcade/bortezumab work in and why?
A
Multiple myeloma
Mantle cell lymphoma.
Two theories;
