What is the multi-hit hypothesis?
A
The theory that cancer is the result of accumulated mutations to a cell’s DNA

What is the evidence for the multi-hit hypothesis?
A
Correlation of age with cancer; if only one mutation was required, most cancers would occur independent of age.
Histologically; can see progression from precursor metaplastic disease to neoplastic eg cervical, barrett’s oesophagus.
Experimentally; myc/ras co-operation in mice
