1
Q
Describe how lysine interacts with ATP
A
- Lys interacts with ATP gamma phosphate, the orientations are diff with ADP
- interacts via salt bridge
- loss of salt bridge causes large rearrangement in 160s loop
2
Q
Lys 161 as an atp sensor
A
- lys can sense atp in nucleotide sensing loop because it can form salt bridges. if atp is there, it will sense the gamma phosphate
3
Q
What does the rearrangement do
A
- rearrangement at interface between beta-hairpin motif and nucleotide sensing loop helps open entrance to the nucleotide-binding site (pocket)
- if have adp , entrance will open
- if have atp, forms a saltbridge and will not allow to have an open entrance
- opening and closing = conformational changes that allow to stabilze your complex
4
Q
motif
A
piece of a tertiary structure and interact to form a domain
5
Q
AMP-PNP
A
ATP mimic that jams reaction so that there cannot be a hydrolysis
