o Can cause headache, flushing, and alter color vision
o Do not take with nitrates or alpha-1 blockers
o MOA: nitric oxide activates guanylate cyclase → increases synthesis of cGMP → cGMP relaxes smooth muscle in the penis → increased blood flow into the penis
• Nitric oxide is released in men when sexually aroused; man must be sexually aroused for these to work
• Nitric oxide is metabolized by phosphodiesterase
• Inhibit PDE → increase cGMP levels → erection lasts longer
